What the study brings together

HER2-LADDER, published in Signal Transduction and Targeted Therapy, combined pretreatment pathology images and clinical variables to study response to neoadjuvant dual HER2-targeted therapy in HER2-positive breast cancer.

H&E images
HoVer-Net analysed cells.
HER2 IHC images
A customized D-PathAI pipeline measured membrane staining intensity and completeness.
Research framework
Image measurements contributed to spatial analysis and response prediction.

The authors call for prospective validation and note potential selection bias in retrospective analyses, alongside variation in staining and scanning.

D-PathAI’s role was image analysis within a separate research framework. The study does not establish a treatment-selection function for the current product.
